588Plasma concentrations of persistent organic pollutants and pancreatic cancer risk

Abstract Background Findings and limitations of previous studies on persistent organic pollutants (POPs) pancreatic cancer risk support conducting further research in prospective cohorts. Objective To investigate the association between plasma concentrations selected POPs risk. Methods Prospective case-control study nested within EPIC cohort. Participants were 513 cases 1020 matched controls. Concentrations 22 (organochlorine pesticides, polychlorinated biphenyls, polybrominated diphenyl ethers) measured collected at baseline by GC-MS/MS. Results Some, generally moderate effects observed higher p,p’-DDT, trans-nonachlor, β-hexachlorocyclohexane, sum 6 organochlorine pesticides 16 POPs. The odds ratio (OR) for upper quartile trans-nonachlor was 1.55 (95% confidence interval 1.06 to 2.26; P trend=0.025). Associations stronger groups predefined as most valid or relevant. Among participants having fasted >6 hours blood collection, ORs significant 10 11 exposure categories. Higher also among with microscopic confirmation than a clinical diagnosis. normal-weight participants, relevant estimates all follow-up ≥10 years, (e.g., trans-nonachlor: OR = 2.14, 1.01 4.53, trend=0.035). Overall, 3 PCBs, 2 sums exposures more clearly associated Conclusions A few moderately increased risks apparent highest certain POPs, sometimes dose-response relationship.
